Zwf encodes glucose-6-phosphate dehydrogenase, which catalyzes the first step of the pentose phosphate pathway (PPP), converting glucose-6-phosphate into 6-phosphogluconate. This pathway supplies both biosynthetic precursors and reducing power in the form of NADPH, essential for anabolic metabolism. The reaction catalyzed by Zwf is frequently the rate-limiting step of the PPP and therefore represents a common target for overexpression in metabolic engineering [276].
Deletion of zwf prevents flux through the oxidative branch of the pentose phosphate pathway [322]. In the absence of this oxidative route, mutant strains can still produce metabolites originating from the pentose phosphate pathway by directing carbon from the Embden–Meyerhof–Parnas (EMP) pathway into the non-oxidative branch of the pentose phosphate pathway. Silencing zwf consequently increases the proportion of carbon channeled through the EMP pathway and the tricarboxylic acid cycle [342].
