The poxB gene encodes pyruvate oxidase, an enzyme that converts pyruvate directly into acetate via a secondary pathway that operates predominantly under aerobic conditions, as it requires oxygen as the terminal electron acceptor [136]. Accordingly, inactivation of poxB in Escherichia coli limits diversion of carbon flux toward acetate formation, thereby conserving carbon for the acetyl-CoA and malonyl-CoA pools to support the biosynthesis of other metabolites [349]. Under anaerobic fermentation conditions, this pathway is strongly attenuated, and deletion of poxB therefore has only a minor effect on overall acetate production [136].
