The aceA gene encodes isocitrate lyase, the key enzyme of the glyoxylate cycle. This enzyme cleaves isocitrate into succinate and glyoxylate, diverting carbon flux away from the tricarboxylic acid cycle. Deletion of aceA abolishes this reaction, preventing isocitrate consumption via the glyoxylate shunt and promoting carbon retention in isocitrate and its upstream tricarboxylic acid cycle intermediates [328]. In contrast, overexpression of aceA enhances flux through the glyoxylate cycle, increasing the supply of pathway intermediates and precursors that can be further converted into downstream derivatives such as glycolate [214].
