The pntAB operon encodes a membrane-bound pyridine nucleotide transhydrogenase (NAD(P)H:NAD⁺ oxidoreductase) in Escherichia coli that catalyzes the transfer of reducing equivalents from NADH to NADP⁺, thereby regenerating NADPH. Overexpression of pntAB has been shown to increase the intracellular NADPH pool, enhancing the synthesis of NADPH-dependent products [348,265].
To maintain an appropriate NADH/NADPH balance, pntAB can be co-overexpressed with the soluble transhydrogenase gene sthA, which catalyzes the reverse transhydrogenation reaction [136].
