tyrA encodes the bifunctional enzyme chorismate mutase/prephenate dehydrogenase, which catalyzes the first two steps of L-tyrosine biosynthesis through distinct catalytic domains, converting chorismate to 4-hydroxyphenylpyruvate via the intermediate prephenate [342, 351]. Both enzymatic activities are subject to feedback inhibition by L-tyrosine [342]. As a rate-limiting step in tyrosine biosynthesis, expression of feedback-resistant variants (tyrAfbr) has been widely employed to enhance intracellular tyrosine availability and improve the production of tyrosine-derived metabolites, including flavonoids, in engineered Escherichia coli [351,360].
