The tdh gene encodes L-threonine dehydrogenase, an enzyme from Escherichia coli that catalyzes the NAD⁺-dependent oxidative dehydrogenation of L-threonine to 2-amino-3-ketobutyrate, a key step in the 2,5-dimethylpyrazine biosynthetic pathway. The E. coli variant has been shown to be more effective than homologs from other strains [358]. Deletion of tdh prevents the degradation of L-threonine, thereby reducing its conversion into downstream metabolites [188].
